Characterization and molecular detection of pathogenicity and antibioticresistance genes in Vibrio parahaemolyticus isolated from pacific white shrimp

ABSTRACT
 
 This study aimed to conduct the characterization and molecular detection of pathogenicity antibiotic-resistance genes in Vibrio parahaemolyticus, as causative agent vibriosis Pacific white shrimp. The V. parahaemolyticus isolates were collected from shrimp’s hepatopancreas, before biochemical test polymerase chain reaction (PCR) sequencing 16S rRNA gene confirmation. hemolysis PCR applied detect presence virulence genes, namely toxR, thermostable direct haemolysin (tdh), tdh-related (trh). Kirby-Bauer method was used for characterizing resistance patterns against ampicillin (AMP), tetracycline (TET), cyprofloxacin (CIP), enrofloxacin (ENR), chloramphenicol (CHL). tests PCR-16SrRNA confirmed that 12 belonged further verified by amplification toxR 382 bp (100% isolates). alpha activity also amplicon 199 all isolates. All showed their AMP 42% TET-resistant. However, no shown CIP, ENR, CHL. PCR-based analysis resulted a detectable ampC (42% isolates) tetB (83% isolates).

 ABSTRAK
 Penelitian ini bertujuan untuk melakukan karakterisasi dan deteksi molekular dari gen patogenisitas resistansi antibiotik pada agen penyebab udang vaname. Isolat dikoleksi hepatopankreas, diuji secara biokimiawi selanjutnya dikonfirmasi dengan (PCR)-sekuensing rRNA. Tes hemolisis metode diterapkan mendeteksi keberadaan virulensi (tdh) Metode Kirby Bauer digunakan pola terhadap ampisilin tetrasiklin kloramfenikol (CHL), siprofloksasin (CIP) enrofloksasin (ENR). Uji biokimia sekuensing memastikan bahwa isolat adalah yang diverifikasi amplifikasi berukuran isolat). Aktivitas alfa juga amplikon (199 bp) di semua isolat.
